(T1030-01-05) GCMS Analysis of Bioactive Compounds of Extracted Growth Hormone Concentration (Auxin) and Antimicrobial Activity of Pentas lanceolata and Chrysanthemum Morifolium: An Ornamental Flowering Plants

Purpose: Traditional medicine is crucial to ethnic groups' their heavy reliance on herbal medicine. Growth hormone, a group of structurally diverse compounds (auxin), are by far the most studied group of plant hormones mainly because they were the first to be discovered. In our study we have taken 2 ornamental plants like Pentas species also called Kabyakyasha. The medicinal properties of pentas species, which are widely used in folk medicine. Chrysanthemum morifolium ‘blossoms of florists' are typically used to make tea, an history of usage in traditional Chinese medicine. The current investigation sought to evaluate the extract powdered sample includes flowers and leaves with phytochemical estimation of methanol solvent extraction for qualitative, quantitative, GCMS, and antimicrobial activity.

Methods: Qualitative analysis of phytochemical: The extract was subjected to preliminary phytochemical assay, to detect the sample was screened for qualitative assay for proteins, carbohydrates, glycosides, tannins, flavonoids, saponins, alkaloids, amino acids, and phenols. Quantitative estimation of phytochemicals: Determination of total phenols- The total phenol concentration in Methanolic leaves and flowers extracts were determined by using Folin-Ciocalteu method. Catechol was used as standard to produce the calibration curve. Determination of total flavonoids- The total flavonoid content of leaves and flowers extracts were measured using aluminium chloride method. Rutin was used as standard for calibration curve. GCMS of bioactive compounds of extracted Growth hormone: Plant materials were collected dried and powdered, then soaked in an organic solvent like ethyl acetate and sample was extracted with GCMS Technique. Sample Preparation: 0.5 g of the powder was taken into a 100 ml conical flask. Added 20 ml of HPLC grade methanol to it. Kept in ultrasonicator for 4 hours. Filtered and injected. Total Run Time: 26.6min, Split Ratio: 10:1, Injection Volume: 2µl, Detector: Mass Detector. Antimicrobial activity: Agar well diffusion method is widely used to evaluate the antimicrobial activity of plants or microbial extracts. Lacto bacillus is inoculated to agar plates and those plates were incubated at 37^O C. The antimicrobial agent diffuses in the agar medium and inhibits the growth of the microbial strain assessed.

Results: Qualitative estimation: The methanol extract of Pentas lanceolata flowers and leaves sample has highest amount of carbohydrates and flavonoids. The methanol extract of Chrysanthemum morifolium leaves have highest concentration of saponins, amino acids, carbohydrates and flowers have amino acid, phenols, carbohydrates, and flavonoids. Quantitative estimation: The methanol extract of Chrysanthemum morifolium has highest portion of bioactive compounds of phenol with standard catechol with compared to Pentas lanceolata. The methanol extract Chrysanthemum morifolium has highest portion of bioactive compounds as flavonoids with standard Rutin with compared to Pentas lanceolata. GCMS of bioactive compounds of extracted Growth hormone: Pentas lanceolata flowers shows list of 18 compounds like Octane, 2,7-dimethyl; Undecane, 3-methyl; Nonane, 3,7-dimethyl; Decane, 2,9-dimethyl; Decane; Octane, 6-ethyl-2-methyl; Nonane; Didodecyl phthalate; 1,2 benzenedicarboxylic acid, diundecyll ester; Phthalic acid, cyclobutyldecyl ester; Octane, 2,2-dimethyl; Octane; Butane, 2,2-dimethyl; 4-Bromo-3-methoxynaphthelene-2-carboxylic acid; 5-[p-Bromophenyl]-2,4-pentadieonic acid ethyl ester; (1,2,4) Triazolo [4,3-a] quinazolin-5(4H)–one, 4-ethyl-1-(trifluoromethyl); 1-Methyl-7-phenyl-3H-[1,3]oxazolo[3,2-g]purine-2,4-dione; and N-[5-(Dimethylamino)naphthalene-1-yl]-2,2,2-trifluoroacetamide. Chrysanthemum morifolium flowers shows list of 9 compounds like Pentadecanoic acid, methyl ester; Tridecanoic acid, methyl ester; Dodecanoic acid, 2-methyl; Cyclopentaneundecanoic acid, methyl ester; Hexadecanoic acid, 14-methyl-, methyl ester; Tetradecanoic acid, 12-methyl-, methyl ester; Dodecanal and Cyclohexane, 1,1`-(2-methyl-1,3-propanediyl)bis. Antimicrobial activity: Antimicrobial activity was observed in gram positive bacteria lactobacillus. The methanol extracts of Pentas lanceolata leaves extract showed MIC (minimum inhibitory concentration) of 2.0 mg/L and flowers sample has 0.6 mg/L. The methanol extract of Chrysanthemum morifolium leaves sample MIC of 1.5 mg/L and flowers sample MIC showed 2.0 mg/L with standard range of ampicillin MIC range from 0.1 mg/L to 0.2 mg/L.

Conclusion: Qualitative estimation: Pentas lanceolata and Chrysanthemum morifolium flowers and leaves have highest concentration of saponins, amino acids, carbohydrates and flowers have amino acid, phenols, carbohydrates, and flavonoids., Quantitative estimation: The extract of Pentas lanceolata and Chrysanthemum morifolium has highest portion of bioactive compounds as flavonoids with standard Rutin. Flavonoids perform many functions like regulating cell growth, attracting pollinators insects, and protecting against biotic and abiotic stresses. GCMS of bioactive compounds of extracted Growth hormone: Pentas lanceolata flowers showed highest peak range with Didodecyl phthalate, showed electroconductive activity.Chrysanthemum morifolium flowers showed the highest peak as compound Pentadecanoic acid, methyl ester, Undecanoic acid, showed antiproliferative and antioxidant activity. Antimicrobial activity: Pentas lanceolata leaves sample showed MIC 2.0 mg/L and Chrysanthemum morifolium leaves and flowers showed MIC range from 1.5 to 2.0 mg/L which infer that both the plants possess antimicrobial activity. We can conclude that bioactive compounds from Growth hormone concentration (auxin) that have various medicinal properties and promising antimicrobial activity.
